What Is SS-31 (Elamipretide)?

SS-31 (also known as Elamipretide or MTP-131) is a mitochondria-targeting tetrapeptide and one of the most potent antioxidant peptides available in Canada. How SS-31 Works

SS-31 concentrates ~1,000-fold at the IMM due to the positive charge of its D-Arg and Lys residues. At the IMM, it intercalates into cardiolipin — the anionic phospholipid essential for cristae architecture and ETC supercomplex organisation. By stabilising cardiolipin’s interaction with cytochrome c, SS-31 maintains ETC Complex III–IV electron transfer efficiency, reduces electron leak and ROS generation, and preserves mitochondrial membrane potential. In aged tissue, where cardiolipin is oxidised and disrupted, SS-31 restores this structural foundation, improving ATP output and reducing oxidative damage.

How does SS-31 get into mitochondria?

SS-31 is cell-permeable and enters mitochondria via its net positive charge (+3), which is attracted to the strong negative mitochondrial membrane potential (~180 mV). Once inside, it concentrates at the IMM by binding cardiolipin’s negatively charged phosphate head groups.

Is SS-31 the same as Elamipretide?

Yes. SS-31 (Szeto-Schiller peptide 31), MTP-131, and Elamipretide are all names for the same D-Arg-Dmt-Lys-Phe-NH₂ tetrapeptide. Elamipretide is the INN name used in study contexts.

How to Reconstitute

Add 1.0 mL of bacteriostatic water to the vial. Inject the water slowly down the inside wall of the vial — do not inject directly onto the powder. Gently swirl until fully dissolved; do not shake. This yields approximately 10.0 mg/mL. Store the reconstituted vial at 4 °C and use within 28 days.

Low Starting Dose

• Dose per injection: 5 mg (5,000 mcg) — 50 units (0.50 mL) on a U-100 insulin syringe
• Frequency: Once daily, subcutaneous injection
• Cycle length: 8–12 weeks
