What Is MOTS-c? Benefits & Overview
MOTS-c is a 16-amino acid mitochondrial-derived peptide (MDP) encoded within the 12S rRNA gene of the mitochondrial genome. Available in Canada from Reviv Peptides as a 10 mg lyophilized vial, MOTS-c regulates metabolic homeostasis via AMPK activation, improves insulin sensitivity, enhances fatty acid oxidation, and has demonstrated anti-aging and exercise-mimetic effects in preclinical models — earning it a position among the most compelling longevity peptides available today.
MOTS-c circulates in human plasma and its levels decline with age, correlating with reduced metabolic flexibility and increased insulin resistance in ageing models.
MOTS-c Benefits & Uses
- AMPK activation and energy sensing pathway research
- Glucose uptake and insulin sensitivity models
- Fat oxidation and energy expenditure studies
- Mitochondrial stress response and retrograde signalling
- Exercise-mimetic and physical performance models
- Ageing and metabolic decline research
- Folate cycle and one-carbon metabolism studies
How MOTS-c Works
Under metabolic stress (glucose deprivation, oxidative stress), MOTS-c translocates from mitochondria to the nucleus where it interacts with AMPK and regulates the folate cycle — particularly the AICAR intermediate in the de novo purine synthesis pathway, which is a direct AMPK activator. AMPK activation increases GLUT4 translocation for glucose uptake, stimulates beta-oxidation, suppresses mTOR-mediated anabolism, and upregulates PGC-1α for mitochondrial biogenesis. In aged mouse models, exogenous MOTS-c restored insulin sensitivity comparable to young controls.

MOTS-c Canada FAQs
How does MOTS-c differ from SS-31 in mitochondrial research?
SS-31 (Elamipretide) is a membrane-targeted peptide that physically stabilises the inner mitochondrial membrane and cardiolipin to improve electron transport chain efficiency. MOTS-c is a signalling peptide that translocates to the nucleus to modulate AMPK and metabolic gene expression. They work at different levels of mitochondrial biology.
Is MOTS-c considered an exercise mimetic?
In preclinical models, MOTS-c injection activates many of the same AMPK-PGC-1α pathways triggered by aerobic exercise, improving metabolic parameters without physical activity — qualifying it as an exercise-mimetic research tool for studies where physical intervention is not possible.

Reconstitution & Dosing Guide
How to Reconstitute
Add 3.0 mL of bacteriostatic water to the vial. Inject the water slowly down the inside wall of the vial — do not inject directly onto the powder. Gently swirl until fully dissolved; do not shake. This yields approximately 3.33 mg/mL. Store the reconstituted vial at 4 °C and use within 28 days.
Low Starting Dose
- Dose per injection: 200 mcg — 6 units (0.06 mL) on a U-100 insulin syringe
- Frequency: Once daily, subcutaneous injection
- Cycle length: 8–12 weeks
Start at the lowest effective dose and adjust based on individual response. Consult a qualified healthcare professional before use.
