What Is KPV? Benefits & Overview

KPV (Lysine-Proline-Valine) is a C-terminal tripeptide fragment of alpha-MSH and one of the most studied anti-inflammatory peptides available in Canada. KPV exerts potent anti-inflammatory effects by directly inhibiting NF-κB, reducing pro-inflammatory cytokines (IL-1β, TNF-α, IL-6), and modulating gut mucosal immunity — with particular promise in models of inflammatory bowel disease, skin inflammation, and systemic immune dysregulation.

Its small size (MW ~341 Da) enables intestinal epithelial cell penetration and nuclear accumulation, where it directly inhibits transcription factors involved in the inflammatory cascade.

KPV Benefits & Uses

  • NF-κB and AP-1 pathway inhibition and inflammation reduction
  • Intestinal epithelial barrier integrity and IBD research models
  • Gut permeability and tight junction protein studies
  • Dermal wound healing and skin repair models
  • Cytokine (IL-1β, TNF-α, IL-6) suppression studies
  • Receptor-independent anti-inflammatory mechanism research

How KPV Works

KPV inhibits NF-κB activation by blocking IκB kinase (IKK) complex activity, preventing p65/p50 nuclear translocation and subsequent transcription of pro-inflammatory genes. This action is entirely independent of melanocortin receptor binding — unlike the parent α-MSH molecule. KPV can enter intestinal epithelial cells via PepT1 transporters, enabling intracellular action at transcription factor level. It also suppresses NLRP3 inflammasome assembly and downstream IL-1β maturation in macrophage models.

KPV Canada FAQs

How is KPV different from BPC-157 for gut research?

BPC-157 drives gut repair via NO, VEGF, and growth factor signalling — primarily angiogenic and structural repair. KPV works upstream by directly inhibiting NF-κB and inflammatory transcription factors. They are complementary tools for IBD models, addressing different layers of the inflammatory-repair cascade.

Does KPV require a receptor to work?

No — KPV’s anti-inflammatory effects are melanocortin receptor-independent. It enters cells via peptide transporters and acts directly on intracellular inflammatory signalling components (IKK, NF-κB, NLRP3).

Reconstitution & Dosing Guide

How to Reconstitute

Add 3.0 mL of bacteriostatic water to the vial. Inject the water slowly down the inside wall of the vial — do not inject directly onto the powder. Gently swirl until fully dissolved; do not shake. This yields approximately 3.33 mg/mL. Store the reconstituted vial at 4 °C and use within 28 days.

Low Starting Dose

  • Dose per injection: 200 mcg — 6 units (0.06 mL) on a U-100 insulin syringe
  • Frequency: Once daily, subcutaneous injection
  • Cycle length: 8–12 weeks

Start at the lowest effective dose and adjust based on individual response. Consult a qualified healthcare professional before use.
